Diese Seminararbeit bietet die M?glichkeit dazu: Die wichtigsten Punkte sind wortw?rtlich ?bersetzt, andere Teile bieten eine ?bersichtliche Zusammenfassung. Die Originalarbeit in englischer Sprache ist in den Universit?tsbibliotheken f?r jeden Medizinstudenten frei zug?nglich. Sialidose ist eine autosomal rezessive Erkrankung, welche durch die gest?rte Funktion von Sailidase Enzymen hervorgerufen wird. Sie wird in zwei Ph?notypen unterteilt, Sialidose Typ I und II, Sialidose ist eine sehr heterogene Erkrankung mit variierenden Alter des Auftretens und Patologien. Derzeit gibt es keine brauchbare Therapie f?r die Behandlung von Sialidose Patienten. Auf molekularer Ebene zeigen die Zellen von Sialidose Patienten die (zusammengesetzte) heterozygote Mutationen aufweisen eine unsachgem??e Enzymfaltung, den Verlust der Aktivit?t des Sialidase Enzyms und deswegen eine Anh?ufung von Sialidase Konjugaten innerhalb der Lysosome. Eine vielversprechende Behandlungsm?glichkeit stellt die Verwendung von kleinen pharmakologischen Molek?len dar, um die enzymatische Aktivit?t der mutierten Proteine zu erh?hen. In dieser Studie untersuchten wir sowohl die Wirksamkeit eines Immunsupressivums (Celastrol) als auch die eines proteosomalen Inhibitors (MG132) um mutierte Enzyme mit alarmierender Konformation zu retten. Unsere Ergebnisse offenbarten, dass MG132 die Enzymaktivit?t erweitert und dass es in Zellen, welche fehlerhafte Sialidase besitzen, lokalisiert ist. Wir fanden au?erdem heraus dass MG123 die Anh?ufung der Gangliosid Produkten GT1b, GD3, GM3 in vorinstallierten Sialidose Zellen reduziert. Andererseits scheint Celastrol die Sialidase Herstellung und Aktivit?t zu reduzieren was einen neuartigen potentiellen Effekt von Celastrol auf Sialidase mit sich bringt. Interessanterweise scheint die Kombination aus Celastrol und MG132 die positive Auswirkung von MG132 auf die k?rpereigene und die rekombinante Expression von besch?digter Sialidase zu verst?rken. <p>Since insulin became available for the treatment of diabetes in 1922 a number of major advances have been made, which include the modification of insulin to vary its timing of action, its purification, and latterly, the production of human insulin. Human insulin in quantities sufficiently large for therapy has been made available by two techniques developed in parallel during the late 1970s. These involve either (i) formulation in E. coli bacteria suitably encoded by DNA recombinant methods of the A- and B-chains of human insulin followed by a chain combination reaction ('biosynthetic' human insulin) or (ii) enzymatic conversion (transpeptidation) of porcine insulin brought to react with a threonine ester by porcine trypsin in a mixture of water and organic solvents, yielding human insulin ('semi-synthetic' human insulin). This book includes the first clinical-pharmacological studies of each of the highly purified 'semi-synthetic' human insulin preparations: Actrapid ? HM; Monotard? HM; Protaphane? HM; Actraphane? HM; and Ultratard? HM (Novo Industri A/S, Copenhagen). The preliminary studies established their safety and efficacy relative to their porcine and bovine counterparts emphasising the relevance of species and formulation on the pharmacokinetics and biological responses to insulin. Additional investigations with human insulin demonstrated the influence of insulin concentration, site of administration, the addition of aprotinin to insulin and the mixing of 'short-' and 'intermediate-acting' formulations on insulin 'bioavailability'.
