Special attention is paid in the book to the systematic analysis of the consequences of the Hodgkin-Huxley mathematical theory of the nervous impulse for the problem of excitability. <p>A preface should justify the existence of the book it precedes and this is invariably done in scientific texts by reference to the explosive growth of the field since the last such volume appeared. In molecular biology, most fields can be justifiably described as growing explosively, as should be the case for a young and vigorous science, but the study of membrane proteins stands out as one which has taken giant strides in the last few years. Ignorance of the structure and function of membrane proteins at the molecular level was certainly not due to lack of interest but rather was a result of lack of appropriate techniques. It has above all been the development of new experimental methods which has wrenched membrane biochemistry out of what Anthony Martonosi fetchingly called its 'romantic phase' (Le. lots of ideas and few facts), into an era when the determination of membrane protein structure and mechanism is a reasonable goal. Membrane proteins are generally classified as peripheral or integral. Peripheral proteins are relatively easily dissociated from membranes by mild treatments whence their study is essentially no different to that of soluble proteins. This book therefore concentrates on integral proteins which are strongly bound to the membrane by hydrophobic interactions with lipids. Membrane is not a good looking or agreeable amoeba. On the whole his character is downright ugly, arrogant and obnoxious. He also stinks, his breath, his armpits, his hair, his ass, his groin, his feet and his pillow all stink. He is in love with his ass-hole and is totally preoccupied with his bowel movements. Membrane is slovenly, lives in filth and is always on the lookout for an easy opportunity to swindle somebody and get the best for himself at the expense of others. Despite this opportunism, Membrane very often comes out as the loser in his adventures and even though they end in a humorous way, there is a kind of pathetic tragedy that comes through that makes the reader think a bit more deeply about the story. <p>The chick embryo chorioallantoic membrane (CAM) is an extraembryonic membrane which serves as a gas exchange surface and its function is supported by a dense capillary network. Because of its extensive vascularization and easy accessibility, the CAM has been broadly used to study the morpho-functional aspects of the angiogenesis process in vivo and to investigate the efficacy and mechanisms of action of pro-angiogenic and anti-angiogenic natural and synthetic molecules. The CAM is a suitable site for transplanting tissues, which can survive and develop in the CAM by peripheral anastomoses between graft and original CAM vasculature or by new angiogenic vessels grown from the CAM that invade the graft. While the formation of peripheral anastomoses between host and pre-existing donor vessels is the main, and the most common, mechanism involved in the revascularization of embryonic grafts, the growth of CAM-derived vessels into the graft is only stimulated in tumor grafts. The CAM has long been a favored system for the study of tumor angiogenesis and metastasis, because at this stage the chick immunocompetence system is not fully developed and the conditions for rejection have not been established. Tumors remain avascular for 72 h, after which they are penetrated by new blood vessels and begin a phase of rapid growth. Also, delivery of tumor cells onto the CAM allows the fine study of the effects of tumor derived angiogenic growth factors on blood vessel structure and functionality. The CAM may also used to verify the ability to inhibit the growth of capillaries by implanting tumors onto the CAM and by comparing tumor growth and vascularization with or without the administration of an anti-angiogenic molecule. Other studies using the tumor cells/CAM model have focused on the invasion of the chorionic epithelium and the blood vessels by tumor cells. <p>With the aim of providing a deeper insight into possible mechanisms of biological self-organization, this thesis presents new approaches to describe the process of self-assembly and the impact of spatial organization on the function of membrane proteins, from a statistical physics point of view. It focuses on three important scenarios: the assembly of membrane proteins, the collective response of mechanosensitive channels and the function of the twin arginine translocation (Tat) system. Using methods from equilibrium and non-equilibrium statistical mechanics, general conclusions were drawn that demonstrate the importance of the protein-protein interactions. Namely, in the first part a general aggregation dynamics model is formulated, and used to show that fragmentation crucially affects the efficiency of the self-assembly process of proteins. In the second part, by mapping the membrane-mediated forces into a simplified many-body system, the dynamic and equilibrium behaviour of interacting mechanosensitive channels is derived, showing that protein agglomeration strongly impacts its desired function.
